Pirolizin—tRNKPil ligaza (EC 6.1.1.26, PylS, pirolizil-tRNK sintetaza) je enzim sa sistematskim imenom L-pirrolizin:tRNKPil ligaza (formira AMP).[1][2][3] Ovaj enzim katalizuje sledeću hemijsku reakciju
U organizmima kao što je metanosarcina barkeri koji inkorporiraju modifikovanu aminokiselinu pirolizin (Pyl) u pojedine metilaminske metiltransferaze, jedana neobična tRNKPyl, sa CUA antikodonom, može direktno da veže pirolizin posredstvom ove klase II aminoacil-tRNK ligaza.
